转录因子HMBOX1在胃癌发生发展中的作用及机制研究的任务书Title：TheRoleandMechanismsofTranscriptionFactorHMBOX1intheOccurrenceandDevelopmentofGastricCancerBackgroundandSignificance:Gastriccancerisoneofthemostcommonmalignanttumorsworldwide,especiallyinAsia.Duetoitscomplexanddiverseetiology,theearlydetectionandinterventionofgastriccancerisofgreatimportanceforpreventingitsfastprogressionandimprovingthesurvivalrateofpatients.Transcriptionfactorsplayacrucialroleinregulatinggeneexpressionandmayinvolveintheinitiationofcarcinogenesis,impactingthebiologicalbehaviorandclinicaloutcomesofgastriccancer.HMBOX1,anewlyidentifiedtranscriptionfactor,isoriginallyidentifiedasahomeobox-containingproteinthatplaysaroleintheregulationofembryonicdevelopment.Inrecentyears,increasingevidencesuggeststhatitalsocontributestothedevelopmentandprogressionofmultiplecancerssuchaspancreaticcancerandbladdercancer.However,theexactroleandmechanismsofHMBOX1ingastriccancerremainlargelyunknown.Thus,itisurgentlyrequiredtoinvestigatethepotentialinvolvementofHMBOX1ingastriccanceranditsunderlyingmechanisms.Objectives:TheaimofthisstudyistoinvestigatetheexpressionandfunctionofHMBOX1ingastriccancer.Specifically,wewill:1.DetecttheexpressionlevelofHMBOX1ingastriccancertissuesandadjacentnormaltissuesbyreal-timePCRandWesternblot,andanalyzethecorrelationbetweentheexpressionlevelofHMBOX1andclinicalpathologicalfeaturesandprognosisofpatientswithgastriccancer.2.InvestigatethebiologicaleffectsofHMBOX1knockdownoroverexpressionontheproliferation,apoptosis,migration,andinvasionofgastriccancercellsinvitro.3.ExplorethepotentialdownstreamtargetsofHMBOX1basedonbioinformaticsanalysisandverifythegenesthataresignificantlyregulatedbyHMBOX1andthesignalingpathwaysthatareinvolvedusinggain-of-functionandloss-of-functionmodels.4.EvaluatetheeffectofHMBOX1ontumorgrowthandmetastasisinvivousingaxenograftmousemodelofgastriccancer.5.InvestigatethemolecularmechanismsunderlyingtheregulationofHMBOX1ingastriccancercells,includingtheexplorationofitspotentialinteractio
