BDNF修饰骨髓间充质干细胞治疗AD的实验研究的开题报告Title:BDNF-modifiedbonemarrowmesenchymalstemcelltherapyforAlzheimer’sdisease:AnexperimentalstudyIntroduction:Alzheimer’sdisease(AD)isaneurodegenerativedisordercharacterizedbytheprogressivelossofcognitivefunctionandmemory.Currently,thereisnocureforAD.However,stemcelltherapyhasemergedasapromisingtreatmentoptionforthisconditionduetothepotentialofstemcellstorepairdamagedtissues.Bonemarrowmesenchymalstemcells(BMSCs)areparticularlypromising,astheycandifferentiateintoavarietyofcelltypesandareeasilyaccessiblefortransplantation.Furthermore,brain-derivedneurotrophicfactor(BDNF)hasbeenshowntoimprovecognitivefunctioninADpatients.Therefore,theaimofthisstudyistoinvestigatethetherapeuticpotentialofBDNF-modifiedBMSCsinthetreatmentofAD.ResearchQuestion:CanBDNF-modifiedBMSCtherapyeffectivelyimprovecognitivefunctioninADrats?Hypothesis:BDNF-modifiedBMSCtherapywillimprovecognitivefunctioninADratscomparedtonon-modifiedBMSCtherapyornotreatment.Methodology:ThestudywillusearatmodelofADinducedbytheadministrationofamyloid-βpeptide.BMSCswillbeisolatedfromthebonemarrowoftheratsandculturedinvitro.TheBMSCswillthenbetransfectedwithaBDNF-expressinglentivirus(experimentalgroup)oracontrollentivirus(controlgroup).Theratswillbedividedintothreegroups:anexperimentalgroupreceivingBDNF-modifiedBMSCs,acontrolgroupreceivingnon-modifiedBMSCs,andashamgroupreceivingnotreatment.CognitivefunctionwillbeassessedusingtheMorriswatermazetest,andtheeffectsonneuronsurvivalandsynapticplasticitywillbeassessedusingimmunohistochemistry,Westernblotting,andelectronmicroscopy.Anticipatedresults:ItisanticipatedthatBDNF-modifiedBMSCtherapywillresultinsignificantimprovementsincognitivefunctioncomparedtonon-modifiedBMSCtherapyornotreatment.Additionally,theremaybeanincreaseinneuronsurvivalandsynapticplasticityinthebrainsofratsreceivingBDNF-modifiedBMSCs.
