SiRNA沉默XIAP基因对骨肉瘤MG-63细胞生物学特性的影响中期报告摘要目的：探讨SiRNA沉默XIAP基因对骨肉瘤MG-63细胞生物学特性的影响。方法：选取人类骨肉瘤MG-63细胞作为研究对象，通过随机分组实验，分别对无处理组、阴性对照组、脂质体对照组和SiRNA处理组进行处理，采用RT-PCR和Westernblotting方法检测XiapmRNA和Xiap蛋白表达水平。采用MTT法检测各处理组的细胞增殖情况，采用AnnexinV-FITC/PI双染尝法检测细胞凋亡率。最后，采用细胞迁移实验和基质侵袭实验检测细胞迁移和侵袭能力。结果：经过SiRNA干预治疗，MG-63细胞中的XiapmRNA和Xiap蛋白表达均得到抑制，同时细胞增殖率随着SiRNA干预治疗时间的延长而减少，细胞凋亡率逐渐增加。此外，SiRNA处理组的细胞迁移和侵袭能力明显下降。结论：SiRNA沉默XIAP基因可以抑制骨肉瘤MG-63细胞增殖，促进细胞凋亡，并且降低细胞迁移和侵袭能力。关键词：SiRNA；XIAP基因；骨肉瘤；生物学特性AbstractObjective:ToinvestigatetheeffectofsiRNA-mediatedsilencingofXIAPgeneonthebiologicalcharacteristicsofosteosarcomaMG-63cells.Methods:HumanosteosarcomaMG-63cellswereselectedastheresearchobject.Randomizedgroupingexperimentwasconductedtotreatnotreatmentgroup,negativecontrolgroup,liposomecontrolgroupandSiRNAtreatmentgroup.RT-PCRandWesternblottingmethodwereusedtodetectXiapmRNAandXiapproteinexpressionlevels.MTTassaywasusedtodetectcellproliferationineachtreatmentgroup,andAnnexinV-FITC/PIdouble-stainingmethodwasusedtodetectcellapoptosisrate.Finally,cellmigrationandinvasionabilityweredetectedbycellmigrationandmatrixinvasionexperiments.Results:AfterSiRNAintervention,theexpressionofXiapmRNAandXiapproteininMG-63cellswasinhibited,andthecellproliferationratedecreasedwiththeprolongationofSiRNAinterventiontime,andthecellapoptosisrategraduallyincreased.Moreover,themigrationandinvasionabilityofSiRNAtreatmentgroupweresignificantlyreduced.Conclusion:SiRNA-mediatedsilencingofXIAPgenecaninhibittheproliferationofosteosarcomaMG-63cells,promotecellapoptosis,andreducecellmigrationandinvasionability.Keywords:SiRNA;XIAPgene;osteosarcoma;biologicalcharacteristics.
