B7-H1在HIV感染患者mDC上的表达及其免疫调节作用研究的开题报告Title：ExpressionofB7-H1onmDCsinHIV-infectedpatientsanditsimmuneregulationroleIntroduction：Humanimmunodeficiencyvirus(HIV)infectionisachronicviralinfectionthatseverelyimpairstheimmunefunctionofinfectedindividuals.ThedepletionofCD4+Tcellsanddysfunctionofdendriticcells(DCs)canleadtoaweakenedimmuneresponseagainstinfectionsandincreasedsusceptibilitytoopportunisticinfections.B7homolog1(B7-H1),alsoknownasprogrammeddeathligand-1(PD-L1),isatransmembraneproteinthatiscommonlyexpressedonvariouscells,includingDCs.B7-H1playsakeyroleinregulatingTcellactivationandimmuneresponse,anditsupregulationhasbeenobservedduringviralinfections.However,theexpressionandroleofB7-H1onDCsinHIV-infectedindividualsremainsunclear.Objective：ThisstudyaimstoinvestigatetheexpressionofB7-H1onmyeloiddendriticcells(mDCs)inpatientswithHIVinfectionanditspotentialimmuneregulatoryfunction.Methods：BloodsampleswillbecollectedfromHIV-infectedindividualsandhealthycontrols.ThefrequencyandexpressionlevelsofB7-H1onmDCswillbeanalyzedbyflowcytometry.ThecytokineprofilesofDCsandTcellswillbemeasuredbyELISA,andtheeffectofB7-H1onDC-Tcellinteractionswillbeassessedbyinvitrococultureassays.Expectedresults：WeexpecttofindanincreasedexpressionofB7-H1onmDCsinHIV-infectedindividualscomparedtohealthycontrols.WealsoanticipatethattheupregulationofB7-H1onmDCsmaycontributetoaregulatoryimmuneresponse,leadingtoimpairedTcellactivation,cytokinesecretion,andreducedimmuneresponsetoHIV.OurfindingsmayprovideabetterunderstandingoftheimmuneregulationmechanismsduringHIVinfectionandaidinthedevelopmentofnovelimmunotherapies.
