Btbd7基因在SD大鼠牙根发育早期的研究中期报告Introduction:Btbd7genemutationshavebeenlinkedtotoothagenesisinhumans.Inthisstudy,weexaminedtheexpressionpatternofBtbd7duringearlytoothdevelopmentintheSprague-Dawley(SD)rat.Methods:RatembryosatvariousdevelopmentalstageswerecollectedandthemandiblesweredissectedforRNAextractionandquantitativereal-timePCRanalysis.InsituhybridizationwasusedtodetectthespatialandtemporalexpressionpatternofBtbd7inmandibularfirstmolartoothgerms.ImmunofluorescenceanalysiswasperformedtodetectthecellularlocalizationofBtbd7protein.Results:OurresultsshowedthatBtbd7mRNAexpressionwasdetectedinthemandibularfirstmolartoothgermsfromembryonicday14(E14)topostnatalday7(P7).ThepeakofexpressionwasobservedatE18andP1.InsituhybridizationandimmunofluorescenceanalysisrevealedthatBtbd7wasprimarilyexpressedindentalpapillamesenchymecells,includingthepreodontoblastsandodontoblasts.Btbd7proteinwasdetectedinthecytoplasmandnucleusofthesecells.Conclusions:OurstudysuggeststhatBtbd7isexpressedindentalpapillamesenchymecellsduringearlytoothdevelopmentinSDratandmayplayaroleintoothmorphogenesis.FurtherstudiesareneededtoinvestigatetheprecisefunctionofBtbd7intoothdevelopment.
