VEGF-C基因修饰的淋巴结移植促进淋巴管再生的初步研究的开题报告Title:APreliminaryStudyonPromotingLymphaticRegenerationbyVEGF-CGeneModifiedLymphNodeTransplantationIntroduction:Lymphaticsystemplaysanimportantroleinmaintainingfluidbalanceandimmunefunctioninthebody.Lymphedema,aconditioncharacterizedbytheaccumulationoflymphaticfluid,isacommoncomplicationincancerpatientswhoundergolymphnodedissection.Currently,thetraditionaltherapyforlymphedemaincludescompressiontherapy,physicaltherapy,andsurgicalintervention.However,thesetherapiesfailtoachievedesirabletherapeuticeffectsduetothelimitedcapacityofthelymphaticsystemtoregenerate.Therefore,noveltherapiesthatcanpromotelymphaticregenerationareurgentlyneeded.VascularendothelialgrowthfactorC(VEGF-C)isacrucialproteinthatplaysasignificantroleinthedevelopmentandmaintenanceofthelymphaticsystem.VEGF-Chasbeenshowntopromotelymphangiogenesis,enhancelymphaticdrainage,andreducetissueedema.Therefore,itisapotentialtherapeutictargetforlymphedematreatment.Hypothesis:WehypothesizethatVEGF-Cgenemodificationcanpromotelymphaticregenerationandimprovelymphedemasymptomsinmicebyenhancinglymphaticvesselgrowthandfunction.WewillinvestigatethishypothesisbyconductingapreliminarystudyofVEGF-Cgene-modifiedlymphnodetransplantation.Methodology:WewilluseamousemodeloflymphedemainducedbylymphnodedissectiontoinvestigatetheeffectofVEGF-Cgene-modifiedlymphnodetransplantationonlymphaticregeneration.Wild-typemiceandVEGF-Cgene-modifiedmicewillbeusedforlymphnodetransplantation.Themicewillbedividedintothreegroups:acontrolgroupreceivingregularlymphnodetransplantation,agroupreceivinglymphnodetransplantationwithVEGF-Cgenemodification,andagroupreceivinglymphnodetransplantationwithsalineinjectionasanegativecontrol.Lymphaticimaging,histologicalanalysis,andWesternblottingwillbeusedtoevaluatetheeffectofVEGF-Coverexpressiononlymphaticvesselgrowth,tissueedema,andlymphedemasymptoms.ExpectedResults:WeexpectthatVEGF-Cgene-modifiedlymphnodetransplantationcanpromotelymphangiogenesis,improvelymphaticdrainag
