RNAi抑制Mx1基因表达对肿瘤VSV溶瘤敏感性的促进作用的中期报告RNAiisatechniqueusedtosilencespecificgenesthroughguidingthedegradationofmRNA.Inthisstudy,weaimedtoinvestigatetheeffectofsilencingMx1geneexpressiononthesensitivityofVSV(VesicularStomatitisVirus)oncolytictherapy.Mx1isainterferon-induciblegenethatencodesaGTPasewhichplaysanimportantroleinantiviralresponse.PreviousstudieshaveshownthathighexpressionofMx1isassociatedwithresistancetoVSVoncolytictherapy.Therefore,wehypothesizedthatsilencingMx1geneexpressioncouldpotentiallyincreaseVSVoncolytictherapyefficacy.Toconductthisstudy,wefirstvalidatedtheeffectofsynthesizedsiRNAtargetingMx1geneontheknockdownofitsexpressioninvitro.ResultsshowedthatthesiRNAeffectivelyreducedtheexpressionofMx1genewhencomparedtocontrolgroup.Next,weevaluatedtheeffectofMx1geneknockdownonVSVoncolytictherapyefficacyinvivousingamurinetumormodel.ResultsshowedthatsilencingMx1geneexpressionsignificantlyincreasedthesensitivityoftumorstoVSVoncolytictherapy.TheaveragetumorsizeintheMx1knockdowngroupwassignificantlysmallerthanthatofthecontrolgroup,suggestingthatreducedMx1expressionenhancedthetherapeuticeffectofVSV.Inconclusion,theresultsofthisstudydemonstratethatsilencingMx1geneexpressioncanenhancethesensitivityoftumorstoVSVoncolytictherapy.Thisfindingcouldhavesignificantimplicationsforthedevelopmentofmoreeffectiveoncolyticvirus-basedtherapiesinthefuture.
