上海交通大学硕士学位论文从催吐萝芙木中提取利血平的工艺研究姓名:孙蕾申请学位级别:硕士专业:微生物与生化药学指导教师:赵凤生20080128硕士学位论文从催吐萝芙木中提取利血平的工艺研究–————————————————————————————————————摘要利血平是一种重要的药用生物碱,有降压功能,作用缓慢而持久。传统的利血平提取工艺以苯为溶剂,提取率较低,且污染严重。本文对传统的提取工艺进行了改进。用含乙醇的酸性水从催吐萝芙木中浸提利血平,用HZ-818大孔树脂纯化,考察了树脂吸附和解吸的条件,并研究了影响利血平结晶的因素。在此基础上,确定了利血平的小试提取工艺:催吐萝芙木根粉经浸提、大孔树脂吸附和洗脱、沉淀生物碱、溶解沉淀、去除胶质、粗结晶、重结晶,制得利血平晶体,利血平纯度达98.4%,提取总收率29.8%。根据小试工艺,在中试规模从催吐萝芙木根粉中提取利血平,形成了一套稳定、高产的提取工艺,连续6批实验的提取总收率为60.6%~74.3%,产品质量符合《中国药典》的规定。这条提取工艺优于传统的苯提取法,适合于工业化生产。关键词:催吐萝芙木,利血平,生物碱,提取硕士学位论文从催吐萝芙木中提取利血平的工艺研究–————————————————————————————————————ABSTRACTReserpine,asignificantalkaloid,actssteadilyandlastinglyforhypertensiontherapy.Traditionalmethodofreserpinepurificationtookbenzeneasextractionsolvent,whichledtoalowrecoveryyieldandsevereenvironmentpollution.Thepaperimprovedthepurificationmethodofreserpine.AcidicwatercontainingethanolwasappliedtoextractreserpinefromRauvolfiavomitoria.HZ-818macroporousresinwasusedtoadsorbreserpineforpurification.Theconditionsofadsorptionanddesorptionwereinvestigated.Moreover,variousfactorsaffectingreserpinecrystallizationwerestudiedandthepurificationprocessinlaboratoryscalewasdetermined.ReserpinewasextractedfromRauvolfiavomitoria,thenitwaspurifiedthroughaprocessincludingadsorptionanddesorptionwithresin,precipitationofalkaloids,dissolutionofprecipitation,sedimentationofgum,crystallizationandrecrystallization.Thepurityofobtainedreserpinewas98.4%.Totalrecoveryyieldwas29.8%.AccordingtothepurificationprocessofreserpinefromRauvolfiavomitoriainlaboratoryscale,pilotexperimentofreserpinepurificationwasstudiedtoachieveastableandhigh-recoveringpurificationprocess.Therecoveryyieldsofsuccessive6experimentswere60.6~74.3%.ThequalityofreserpinewascheckedaccordingtoChinesePharmacopoeiaandwasuptostandards.Thepurificationprocessissuperiortothetraditionalpurificationprocesswithbenzene.Itissuitabletoindustrialproductionofreserpine.Keywords:Rauvolfiavomitoria,reserpine,alkaloid,purifica
