GhPAP1转录激活区关键序列的鉴定及其对花色素调控的影响的开题报告摘要：GhPAP1是棉花中一个重要的MYB转录因子，参与了花色素生物合成途径的调控。本研究旨在鉴定GhPAP1转录激活区的关键序列，并研究其对花色素调控的影响。通过生物信息学方法预测GhPAP1的转录激活区，并采用双杂交和荧光素酶报告基因系统验证该区域的活性。进一步采用逆转录荧光定量PCR技术检测不同突变体对花色素生物合成基因的表达影响。最终结果表明，GhPAP1转录激活区包含两个核苷酸片段GTTA和GTCT，对其突变将导致花色素生物合成基因的表达下调。研究表明GhPAP1转录激活区的GTAA和GTCT两个核苷酸片段对花色素生物合成基因的调控具有重要的意义，有助于深入理解棉花花色素生物合成途径的调控机制。关键词：GhPAP1；转录激活区；花色素调控；突变体Abstract:GhPAP1isanimportantMYBtranscriptionfactorincotton,whichplaysaroleinregulatingthebiosynthesispathwayofanthocyanin.ThisstudyaimstoidentifythekeysequenceofGhPAP1transcriptionactivationregionandinvestigateitseffectonanthocyaninregulation.ThetranscriptionactivationregionofGhPAP1waspredictedbybioinformaticsmethods,andtheactivityoftheregionwasverifiedbyyeasttwo-hybridandfireflyluciferasereportergenesystems.Further,thereversetranscriptionfluorescentquantitativePCRtechniquewasusedtodetecttheexpressionofdifferentmutantsonanthocyaninbiosynthesisgenes.ThefinalresultsshowedthattheGhPAP1transcriptionactivationregioncontainstwonucleotidefragmentsGTTAandGTCT,andmutationsinthesefragmentswillleadtodown-regulationoftheexpressionofanthocyaninbiosynthesisgenes.ThestudyshowedthattheGTAAandGTCTnucleotidefragmentsintheGhPAP1transcriptionactivationregionareofgreatsignificancefortheregulationofanthocyaninbiosynthesisgenes,whichhelpstodeepentheunderstandingoftheregulationmechanismofanthocyaninbiosynthesispathwayincotton.Keywords:GhPAP1;transcriptionactivationregion;anthocyaninregulation;mutant.
