KLF15对AdipoR1表达的转录调控研究的开题报告Title:TranscriptomeAnalysisofKLF15-regulatedAdipoR1ExpressionBackground:Adiponectinreceptor1(AdipoR1)isatransmembraneproteinthatplaysacriticalroleintheregulationofglucoseandlipidmetabolism.TheexpressionofAdipoR1isregulatedbyvarioustranscriptionfactors,includingKrüppel-likefactor15(KLF15),whichishighlyexpressedinadiposetissueandskeletalmuscles.PreviousstudieshaveshownthatKLF15regulatesAdipoR1expression,butthepreciseregulatorymechanismsremainunclear.Objective:TheaimofthisstudyistoinvestigatetheregulatorymechanismsofKLF15onAdipoR1expressionusingtranscriptomeanalysis.Method:Wewillcollectadiposetissueandskeletalmusclesamplesfromwild-typemiceandKLF15knockoutmice.TotalRNAwillbeextracted,andRNAsequencingwillbeperformedtoanalyzethetranscriptomeprofilesofthesamples.Differentialexpressionanalysiswillbeconductedtoidentifygenesthataredifferentiallyexpressedbetweenthetwogroups.Functionalenrichmentanalysiswillbeperformedtoexplorethebiologicalpathwaysthatareassociatedwiththedifferentiallyexpressedgenes.TheexpressionlevelsofcandidategeneswillbevalidatedbyqPCR.ExpectedResults:Weexpecttoidentifygenesthataredifferentiallyexpressedbetweenwild-typemiceandKLF15knockoutmice.Functionalenrichmentanalysiswillrevealthebiologicalpathwaysthatareassociatedwiththedifferentiallyexpressedgenes.Furthermore,ourstudywillprovideinsightsintotheregulatorymechanismsofKLF15onAdipoR1expression.Conclusion:OurstudywillprovideabetterunderstandingofKLF15-regulatedAdipoR1expressionandwillcontributetothedevelopmentofnewtherapeuticstrategiesformetabolicdisorderssuchasobesityandtype2diabetes.
